GHK-Cu is a naturally occurring copper complex of the tripeptide glycyl-L-histidyl-L-lysine bound to copper(II), first isolated in 1973 by Dr. Loren Pickart. It is naturally present in human plasma, saliva, and urine, and declines roughly 50 percent or more after age 60.

The clinical hypothesis: restoring GHK-Cu concentrations via topical or systemic administration can slow collagen loss and accelerate tissue repair.

How GHK-Cu Works

GHK-Cu's mechanism operates through parallel pathways. Different applications target different pathways.

MMP Modulation and Collagen Remodeling

MMPs (matrix metalloproteinases) break down extracellular matrix. GHK-Cu has a biphasic relationship: at physiological concentrations it drives healthy remodeling. At excessive concentrations it can tip into net collagen fragmentation. That biphasic response is the mechanism behind the copper uglies. It is not a sign the product is not working.

Fibroblast Activation and Collagen Synthesis

Directly stimulates dermal fibroblasts to produce collagen types I and III, elastin, and glycosaminoglycans. Liposomal formulations show 40 to 60 percent greater improvements versus conventional formulations. The carrier system matters as much as the labeled concentration.

Anti-Inflammatory Signaling

Suppresses TNF-alpha and IL-1 beta. Downregulates NF-kB. Explains the post-procedure utility. The anti-inflammatory effect is systemic when injected, which is the rationale for injectable in tissue repair. Human clinical evidence for the injectable application is mostly animal models so far.

Hair Follicle Stimulation

Enlarges follicle size, stimulates keratinocyte growth factor, extends the anagen phase. Real and documented. Does not involve DHT or 5-alpha reductase inhibition. Not a DHT blocker. Should not replace finasteride in male pattern hair loss.

Copper uglies phase: accelerated cellular turnover brings congestion, dullness, small bumps. Do not stop. Reduce to every other day if pronounced. Starting at 0.1 percent topical for the first two weeks reduces this effect significantly.

  • Replacing finasteride for hair loss

My skin looks worse 10 days in. Did I make a mistake?

Probably not. You are likely in the copper uglies window. Drop frequency to every other day, hydrate, and protect from sun. Most users see the skin pivot toward improvement between week 3 and week 5. If you suspect true copper sensitivity (worsening through week 6 with no improvement), stop and discuss with your physician.

Topical or injectable?

Topical has human RCT support including the 2024 multicenter post-procedure study. Injectable is appropriate for systemic tissue repair under physician supervision but does not have the same cosmetic evidence base. If your goal is visible skin improvement, start topical.

Will GHK-Cu help my hair loss?

It enlarges follicles, stimulates keratinocyte growth factor, and extends the anagen phase. It is not a DHT blocker. Use it alongside finasteride, not instead of it, for male pattern hair loss.

Can I mix GHK-Cu into my regular skincare?
